Expression detection and partial cloning of porcine leptin receptor (OBR) gene

The product of the obesity gene, called leptin, is an important regulator of adiposity, which mainly functions via its regulation of feed intake and energy metabolism. Both obesity gene (ob gene) and leptin receptor gene (OBR gene) are thought to play a major role in controlling of body fat mass as well as body weight. The result of RT-PCR shows that levels of pig OBR mRNA are higher in hypothalamus, lung and liver, and lower expression can be detected in other tissues. Total RNA purified from 11 different organs and tissues have been hybridized with pig OBR cDNA probes. The hybridization signals are shown in 7 organs and tissues. 4.1 and 3.8 kb bands were observed from hypothalamus, whereas 3.8 and 3.5 kb bands were observed in other tissues instead. The nearly complete sequence of the extracellular domain of pig OBR gene was obtained. The homology of sequence is 89.2% between pig and human, 80.3% between pig and mouse. 鸡生长激素基因单核苷酸多态与生长及屠体性状的相关性被引量:36

以鸡生长激素(GH)基因作为控制鸡生长和屠体性状主基因的候选基因,以明星肉鸡和丝毛乌骨鸡杂交产生的F_2代鸡群为实验材料,通过PCR-RFLP方法对GH基因进行多态性检测。在该基因内含子3中发现1处碱基突变:G→A,由内切酶EcoR V酶切证实;在内含子4中发现1处碱基突变:C→T,由内切酶Msp I酶切证实,并由此对F_2代鸡群个体进行基因型鉴定。方差分析结果显示,在内含子3中的碱基突变与腹脂性状显著相关;内含子4中的碱基突变与腹脂、胸肉屠体性状显著相关,但两处突变与其他大部分生长和屠体性状相关不显著,结果表明鸡GH基因可能是控制某些屠体性状的主基因,或与控制这些性状的主基因紧密连锁。

多趾是脊椎动物常见的肢体异常,在不同物种间具有相似的表型。研究表明,有相同的基因和发育机理控制不同物种间的多趾表型;最近在人和鼠上的研究进一步表明PPD应是由于干扰了位于Lmbrl内含子内的Shh长程顺式调控元件引起的。对脊椎动物多趾性状的发生机理和相关基因的研究进展进行了综述。

Insulin-like growth factor 2 as a candidate gene influencing growth and carcass traits and its bialleleic expression in chicken被引量:5

We have identified DNA polymorphisms in the gene of insulin-like growth factor 2 by PCR-SSCP in a resource population, which was generated by Silky reciprocally crossing to Broilers. A C→G mutation was detected in the exon 2 (at position 71) by sequencing. This single nucleotide polymorphism (SNP) was found to be associated with production traits. Chicken with BB genotype showed more chest angle width but less 3 week body weight and glandular stom- ach weight than chicken with AA genotype (P<0.05); while the heterozygote (AB genotype) chicken had more abdominal fat weight, eviscerated yield with giblet than AA homozygote chicken. Further analysis showed that there were different genetic effects on some traits between heterozygote AB (paternal allele given first) and heterozygote BA: chickens with genotype BA had more birth weight and breast weight but less abdominal fat weight than chickens with geno- type AB (P<0.05), which could be hypothetically contributed by genome imprinting. Therefore, Silky chickens were selected for production of heterozygotes to confirm whether IGF2 locus was imprinting. Progeny from heterozygote × homozygote reciprocal cross was assayed for ex- pression after the genotype was determined. The transcription of IGF2 was detected by RT-PCR-SSCP. IGF2 gene was expressed bialleleically in 1-day-old neonatal liver and 90-day-old liver, kidney, heart, and muscle of both heterozygote AB and BA chickens. Therefore, IGF2 was not an imprinting gene in chicken. The different genetic effects between the het- erozygote AB and BA remain to be elucidated.

基于F-2群体的鸡重要生长性状遗传分析被引量:22

采用F-2设计,以丝羽乌骨鸡(C系)为一亲本,分别与农大褐蛋鸡(B系),及法国明星肉 鸡(A系)进行正反交,产生了包含A × C,C × A;B × C,C × B 4个杂交组合的F2代群体,建 立了可用于定位产肉及产蛋等性状QTL的资源群体。基于这一F-2群体,对体重及日增重等 性状表型值进行分析,结果显示,亲本间均值差异显著,分离群体变异大,这组性状间有中等 或较高的相关,各性状潜在的QTL座位数不超过10,所建资源家系能够满足QTL定位所需的 样本数量,达到了预期的效果。

Single nucleotide polymorphism analysis on chicken extracelluar fatty acid binding protein gene and its associations with fattiness trait被引量:7

Fattiness is an important parameter to estimate meat quality, which has high heritability. In this experiment, F2 chickens derived from Broilers crossing to Silky were used to study the effect of extracellular fatty acid binding protein (EX-FABP) gene on abdominal fat accumulation. 1.6 kb of the 5′ region of the gene was amplified by six pairs of primers, and then single nucleotide poly-morphisms (SNPs) were detected by the technique of single strand conformation polymorphism (SSCP) and then confirmed by sequencing. There were four nucleotides variations found, A-G at -1807, C-A at -1805, T-C at -1011 and a C insertion at -1000 respectively. The result of least square analysis suggests that the birds with BB genotype defined by the second pair of primer have a higher abdominal fat weight and abdominal fat percentage than the birds with the other genotypes (AA and AB). It implied that EX-FABP gene could be a candidate locus or linked to a major gene to significantly affect abdominal fat traits in chicken.

雌激素受体基因(ESR)是控制猪高产仔数的主效基因之一,具有明显的基因效应:BB型比AA型母猪胎总产仔数和产活仔数分别高出1 40~3 37和0 63~3 58头,是目前商品猪育种和生产中重点检测的主要基因之一。常规采用PCR -RFLPs方法区分该基因由点突变造成的3种不同的基因型。本研究建立1种基于PCR的SSCP(single-strand edconformationpolymorphism)方法对猪ESR该位置的点突变进行检测,具有操作简便、灵敏度高和不需要酶切等优点 。

中国部分地方鸡种B-LⅡβ(β_1外显子)基因分子遗传多态性研究被引量:23

应用PCR-RFLPs方法对10个鸡种的B-LⅡβ(β1外显子)基因进行分子遗传多态性研 究,综合4种限制性内切酶的酶切情况,共检测到37种基因组合型,并且在个体间以及品种间 的基因型频率、基因组合型频率都存在较大的差异。同时各酶切位点的Hardy-Weinberg平衡状态在品种上也表现不一致。克隆测序结果表明:β1外显子分子遗传多态性更多地体现在氨 基酸水平,作为抗原结合区,其丰富的多态性与抗原多样性相一致。
