Carboxymethylpachymaran Enhances Immunologic Function of Dendritic Cells Cultured in Two Kinds of Hepatoma Carcinoma Cell Line's Supernatant via Nuclear Factor κ B/Rel Pathway被引量:9

Objective:To study the immunologic function of dendritic cells(DCs) cultured in two kinds of hepatoma cell line's supernatant and the enhancing effects of carboxymethylpachymaran(CMP) on DCs.Methods:DCs were harvested after stimulation by granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin(IL)-4 from umbilical cord blood using density-gradient centrifugation method. Cultured supernatant of two hepatoma cell lines(HepG2 and HepG2.2.15) were collected for condition medium(CM) according to a volume ratio of supernatant to incomplete RPMI-1640 medium,which was 3:1.CMP was dissolved in incomplete RPMI-1640 medium.Experimental groups were divided according to the culture medium,either CM or with CMP in it.DCs subsets CD83,CD86,CD1a,and d-related human leukocyte antigens(HLA-DR) were analyzed by flow cytometry.The proliferation ability of allogeneic T cells in mixed lymphocyte reaction(MLR) stimulated by DCs was examined by 3-(4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide(MTT) analysis.IL-12p70,interferon-γ(IFN-γ),and nuclear factor k B(NF- k B) were detected by enzyme-linked immunosorbent assay analysis.Results:The proliferation of lymphocytes and secreting level of IL-12 and expression of phenotype of DCs cultured in two kinds of CM were lower than those of normal group(P<0.01).Compared with the normal group,groups treated with CMP showed a higher expression level of DCs subsets,lymphocyte reproductive activity,as well as IL-12 and IFN-γsecretion levels.Groups treated with CMP also demonstrated higher levels of DCs phenotype expression and IL-12 and IFN-γsecretion in supernatant of MLR and higher lymphocyte reproductive activity compared with CM group(P<0.05).Compared with the normal group,the expression level of NF-k B in DCs nuclear was higher in CMP groups but lower in two CM groups(P<0.05).After CMP was added,the NF-κB expression levels of two CM groups were increased compared with levels before CMP was added(P<0.05).However,there was no significant difference between the tw

Andrographolide as An Anti-H1N1 Drug and the Mechanism Related to Retinoic Acid-Inducible Gene-I-Like Receptors Signaling Pathway被引量:9

Objective:To observe the anti-virus effects of andrographolide(AD) on the retinoic acid-inducible gene-l(RIG-l)-like receptors(RLRs) signaling pathway when immunological cells were infected with H1N1.Methods:Leukomonocyte was obtained from umbilical cord blood by Ficoll density gradient centrifugation,and immunological cells were harvested after cytokines stimulation.Virus infected cell model was established by H1N1 co-cultured with normal human bronchial epithelial cell line(16HBE).The optimal concentration of AD was defined by methyl-thiazolyl-tetrazolium(MTT) assay.After the virus infected cell model was established,AD was added into the medium as a treatment intervention.After 24-h co-culture,cell supernatant was collected for interferon gamma(IFN-γ) and inteiieukin-4(IL-4) enzyme-linked immunosorbent assay(ELISA) detection while immunological cells for real-time polymerase chain reaction(RT-PCR).Results:The optimal concentration of AD for anti-virus effect was 250 μg/mL IL-4 and IFN-γ in the supernatant and mRNA levels in RLRs pathway increased when cells was infected by virus,RIG-I,IFN-β promoter stimulator-1(IPS-1),interferon regulatory factor(IRF)-γ,IRF-3 and nuclear transcription factor κB(NF-κB) mRNA levels increased significantly(P<0.05).When AD was added into co-culture medium,the levels of IL-4 and IFN-γ were lower than those in the non-interference groups and the mRNA expression levels decreased,RIG-I,IPS-1,IRF-γ,IRF-3 and NFk B decreased significantly in each group with significant statistic differences(P<0.05).Conclusions:The RLRs mediated viral recognition provided a potential molecular target for acute viral infections and andrographolide could ameliorate H1N1 virus-induced cell mortality.And the antiviral effects might be related to its inhibition of viral-induced activation of the RLRs signaling pathway.

人脐带血来源树突状细胞、巨噬细胞和单核细胞体外培养及鉴定被引量:5

目的建立从人脐带血体外诱导扩增树突状细胞、巨噬细胞和单核细胞的方法。方法从正常人脐带血中分离出单核细胞,经3种细胞因子—重组人粒细胞-巨噬细胞集落刺激因子(rh GM-CSF)、重组人白细胞介素4(rh IL-4)和重组人肿瘤坏死因子(rh TNF-α)联合诱导培养,7 d后收获细胞,并利用光学显微镜、免疫组化的方法进行鉴定。结果培养收获了高纯度的树突状细胞、巨噬细胞、单核细胞,细胞高水平表达CD1a、CD86、CD68和CD14。结论人脐带血单核细胞经细胞因子诱导培养可以得到大量的成熟树突状细胞、巨噬细胞和单核细胞。

野生型PTEN诱导卵巢癌SKOV3细胞生物特性改变

【目的】探索野生型PTEN基因对卵巢癌细胞生物学特性的影响。【方法】将野生型PTEN基因克隆到pAdxsi腺病毒载体并感染SKOV3细胞。荧光镜下检测腺病毒载体对SKOV3细胞的感染效率;用CCK-8法检测细胞增殖的抑制效率;RT-PCR与WB分别检测PTEN mRNA与蛋白表达水平变化;免疫化学法与间接荧光法检测PTEN在细胞内的定位及细胞内NEDD4-1和RAD51等分子表达。【结果】稳定转染PTEN后,24h内PTEN分布于细胞质与核内,而72h后则主要集中于细胞核内;显著增加细胞质内NEDD4-1和细核RAD51等分子表达。【结论】野生型PTEN可影响SKOV3细胞增殖,并诱导细胞表达RAD51与NEDD4-1分子,有利于细胞DNA修复与细胞增殖抑制。

澳门地区产超广谱β-内酰胺酶大肠埃希菌基因型分析被引量:4

【目的】了解澳门地区医院感染产超广谱β-内酰胺酶(ESBL)大肠埃希菌的临床分布特点和基因型特征。【方法】收集2007年12月至2008年2月期间从澳门地区两家医院的临床患者中分离到的大肠埃希菌158株,用表型确定试验确定产ESBL大肠埃希菌。应用PCR方法分别扩增产酶菌株的CTX-M-3、CTX-M-9、TEM和SHV等常见的β-内酰胺酶基因,并结合临床分离菌株的分布特点对PCR扩增结果作进一步的分析。【结果】澳门地区临床分离的158株大肠埃希菌中,有54株产ESBL,产酶率为34.2%。PCR扩增结果显示,所有产酶菌株均携带TEM基因;其中有47株(87%)同时携带CTX-M-9、TEM两个基因;没有菌株携带CTX-M-3和SHV基因。产ESBL大肠埃希菌广泛分布于临床各科室,其中普外科占40.7%,内科占27.8%,心脏科占16.7%。标本分布较为集中,尿液占55.6%,分泌物或脓液标本占22.2%,血液和呼吸道标本各占11.1%。【结论】澳门地区医院感染中,产ESBL大肠埃希菌在临床各科室分布广泛,主要分离自尿液、分泌物或脓液标本;所有菌株均携带TEM基因,并有87%的菌株同时携带CTX-M-9基因。

心力衰竭患者Copeptin、BNP、NT-proBNP、ANP分子的检测及其临床意义被引量:4

目的观察心力衰竭患者血浆和肽素(Copeptin)、ANP、BNP与NT-proBNP水平变化,探讨检测指标与心功能的关系。方法连续选择2012年3月1日~2013年3月31日本院心内科收治的102例心力衰竭患者(心衰组),根据纽约心脏协会(NYHA)心功能分级标准,分为心功能Ⅱ级31例,Ⅲ级36例,Ⅳ级35例,以同期住院心功能正常的患者50例作为对照组。所有患者均空腹抽取静脉血,采用酶联免疫吸附测定法(ELISA)测定血清CRP;自动生化仪测定总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、空腹血糖(FPG)等相关生化指标;Copeptin、BNP、NT-proBNP与ANP等水平应用化学发光法检测;应用SPSS 13.0统计软件分析各指标间的关系。结果①心衰患者血浆Copeptin、BNP、NT-proBNP、ANP水平显著高于对照组(P〈0.01)。在心衰组中,血浆Copeptin水平在心功能Ⅳ级明显高于心功能Ⅲ级患者,心功能Ⅲ级高于心功能Ⅱ级,各组间差异均有统计学意义(P〈0.05)。②心衰患者血浆Copeptin与BNP、NT-proBNP、ANP呈明显正相关。结论心衰患者血浆Copeptin、BNP、NTproBNP、ANP水平随NYHA分级逐级增高,且与心衰严重程度有良好的相关性,Copeptin、BNP、NT-proBNP和ANP水平均可作为临床诊断心衰、评估心功能的敏感检测指标。

参苓散调节小鼠肠道菌群作用的实验研究被引量:22

目的观察参苓散对小鼠肠道菌群的影响。方法 48只雄性昆明鼠,随机分为4组,即低、中、高3个剂量组和对照组,连续给予参苓散14 d,实验前后检测肠道乳杆菌、双歧杆菌、肠杆菌、肠球菌的数量。结果中剂量组的小鼠肠道内双歧杆菌显著增加,肠杆菌减少,乳杆菌和肠球菌无显著变化;高剂量组的小鼠肠道内的双歧杆菌和乳酸杆菌的数量显著增多,肠杆菌的数量显著降低,肠球菌无变化。结论参苓散具有调节小鼠肠道菌群及促进双歧杆菌、乳杆菌增殖的功效。

海洋放线菌素X2对CVB3感染ECV304细胞VCAM-1表达的影响

目的研究柯萨奇病毒(Coxsackievirus,CVB3)感染ECV304细胞后,血管细胞黏附分子-1(vascular cell adhesionmolecule,VCAM-1)的表达变化及海洋放线菌素X2对CVB3感染ECV304细胞VCAM-1表达的影响。方法采用MTT法检测不同浓度的海洋放线菌素X2作用病毒后的抑制情况;采用RT-PCR和FCM分别测定CVB3感染的ECV304细胞在海洋放线菌素X2作用前后不同时间点的VCAM-1的mRNA和蛋白的表达水平。结果海洋放线菌素X2在病毒吸附前对CVB3的SI为63.88。正常状态下ECV304细胞基本无VCAM-1 mRNA表达,CVB3感染ECV304细胞促进VCAM-1 mRNA表达,感染12 h达到最高峰,6~54 h时VCAM-1 mRNA水平,与细胞对照组相比CVB3感染组差异有统计学意义(P<0.05)。CVB3感染ECV304细胞后,VCAM-1蛋白表达在12~48 h显著高于正常ECV304细胞对照组(P<0.05)。海洋放线菌素X2干预后,于12~54 h降低ECV304细胞VCAM-1 mRNA的水平,12~24 h降低VCAM-1蛋白的表达,与CVB3感染组相比差异有统计学意义(P<0.05)。结论海洋放线菌素X2在病毒吸附前对CVB3具有抗病毒作用,并可下调CVB3诱导的ECV304细胞VCAM-1的mRNA水平和蛋白的表达。

Effect of modified Taohongsiwu decoction on patients with chemotherapy-induced hand-foot syndrome被引量:17

OBJECTIVE:To observe the effect of the Traditional Chinese Medicine,modified Taohongsiwu decoction,versus pyridoxine on patients with hand-foot syndrome(HFS)from capecitabine,sorafenib,and gefitinib chemotherapy for gastric,lung,breast,colon,or rectal cancer.Also,to compare quality of life of patients in each group.METHODS:Patients were assigned randomly to group A or B.Group A was given modifiedTaohongsiwu decoction to soak hands and feet for 30 min,once daily.Group B was given 100 mg pyridoxine orally,twice daily.After a 2-week treatment,thetherapeutic effect was assessed by observing three major symptoms,including pain,ulceration,and muscular atrophy.This was assessed with the HFS-14 questionnaire.RESULTS:Significant differences were observed between the two groups in pain relief,and improvement of daily life,walking,and interpersonal communication(P<0.01).No significant differences in driving ability or interpersonal relationships were found.After 2 weeks,the effective rate was 88.3%in group A,which was significantly higher than the50%in group B(P=0.00).CONCLUSION:Modified Taohongsiwu decoction is effective in the treatment of patients with HFS.It improves patients'quality of life according to the HFS-14.
