Folic acid-conjugated liposomal vincristine for multidrug resistant cancer therapy被引量:3

We encapsulated vincristine into folic acid-conjugated PEGylated liposomes to improve the anti-tumor efficacy on multidrug resistant cancers.It was observed that the drug delivery system we constructed exhibited maximum cytotoxicity on KBv200 cells(multidrug resistant variant)compared with any other formulations.The semi-quantitative analysis of region of interest revealed that there was a great increase in area under curve(AUC)of a near-infrared fluorescein in solid tumors due to folic acid-mediated accumulation.Folic acid-conjugated PEGylated liposomes showed a significant tumor growth inhibiting effect in vitro and in vivo.TUNEL assay revealed that folic acid-conjugated PEGylated liposomes could induce cell apoptosis much more greatly than others.This study demonstrated that it had potential application prospective for the treatment of multidrug resistant cancer.

Overcoming multidrug resistance(MDR) in cancer by nanotechnology被引量:5

The emerging nanotechnology-based drug delivery holds tremendous potential to deliver chemotherapeutic drugs for treatment of multidrug resistance(MDR) cancer.This drug delivery system could improve the pharmacokinetic behavior of antitumor drugs,deliver chemotherapeutic drugs to target sites,control release of drugs,and reduce the systemic toxicity of drugs in MDR cancer.This review addresses the use of nanotechnology to overcome MDR classified on the bases of the fundamental mechanisms of MDR and various approaches to deliver drugs for treatment of MDR cancer.

镜像配基筛选:克服“酶屏障”的多肽、寡核苷酸药物研发技术

生物体内存在的"酶屏障"是功能性多肽和寡核苷酸成药的重要瓶颈.以镜像噬菌体展示技术(mirror-image phage display)和镜像适配体筛选技术(Spiegelmer technology)为代表的镜像配基筛选技术能有效地克服这一屏障,获得对靶点具有特异识别能力且在生理条件下高度稳定的D型多肽和L型寡核苷酸配基,有望成为多肽或寡核苷酸类特异性药物研发的有利工具.本文主要综述镜像配基筛选技术的原理及其在药学领域的研究进展.

Nanocarriers for siRNA delivery to overcome cancer multidrug resistance被引量:1

Multidrug resistance(MDR)is an extremely complexed phenomenon in tumor chemotherapy and is one of the major obstacles for successful treatment.Discovery of RNA interference(RNAi)offers a new strategy with great potential to reverse MDR.Specific genes,which contribute to the emerging of MDR,can be silenced by RNAi.However,many natural barriers have to be overcome for efficient and safe delivery of siRNA.In recent years,the various kinds of nanocarriers,such as liposomes,cationic polymers,and inorganic materials,have been developed to deliver siRNA and show good results in reversing MDR.This review mainly summarizes the barriers in siRNA delivery and recent progress in designing nanotechnology-based siRNA delivery systems to overcome tumor MDR.
