Preparation and characterization of doxorubicin functionalized tiopronin-capped gold nanorods for cancer therapy被引量:1

Gold nanomaterials are immerging candidates in medical diagnosis and treatment.Among them,gold nanorods(Au NRs)are widely used for cancer treatment.Tiopronin as a novel thiol drug was used to stabilize Au NRs in this work.Doxorubicin(DOX),a chemotherapeutic drug which works by interacting with DNA to arrest the cell cycle and induce apoptosis,was linked to Au NRs through electrostatic reaction with tiopronin,to obtain Au-TIOP-DOX NRs.Au NRs are also regarded as hyperthermia agents for photothermal cancer treatment.This delivery system(Au-TIOP-DOX NRs)was designed for passively targeting tumor cells in cancer therapy.More importantly,the carboxyl groups of tiopronin can be modified with some biological molecules(DNA/RNA,peptides,or drugs)to make Au NRs as a novel drug-delivery system for cancer treatment.

Fullerenes as unique nanopharmaceuticals for disease treatment被引量:3

As unique nanoparticles,fullerenes have attracted much attention due to their unparalleled physical,chemical and biological properties.Various functionalized fullerenes with OH,NH2,COOH,and peptide modifications were developed.It summarized the biological activities of fullerenes derivatives in cancer therapy with high efficiency and low toxicity,as reactive oxygen species scavenger and lipid peroxidation inhibitor,to inhibit human immunodeficiency virus and to suppress bacteria and microbial at low concentration.In addition,the mechanism for fullerene to enter cells and biodistribution of fullerene in vivo was also discussed.This research focuses on the current understanding of fullerenes-based nanomaterials in the potential clinical application as well as biological mechanism of fullerenes and its derivatives in disease therapy.

AlO(OH)纳米片和纳米棒的水热法制备及其对牛血清白蛋白的吸附作用被引量:1

以EDTA-2Na为螯合剂,铝盐为铝源,尿素为沉淀剂,采用水热法并使反应物以气液界面接触反应合成勃姆石型[AlO(OH)]纳米片及纳米棒;以透射电子显微镜(TEM)、X射线衍射仪(XRD)、傅里叶红外光谱仪(FTIR)等仪器对样品的尺寸、相位、形貌等进行了表征。结果表明,反应温度、时间等对样品的形貌、结晶度等起决定作用,合成的AlO(OH)对牛血清白蛋白(BSA)有较高的吸附能力,且在Al与BSA的质量比为1∶2时,吸附效果最佳。

pH-responsive mesoporous silica nanoparticles employed in controlled drug delivery systems for cancer treatment被引量:8

In the fight against cancer, controlled drug delivery systems have emerged to enhance the therapeutic efficacy and safety of anti-cancer drugs. Among these systems, mesoporous silica nanoparticles(MSNs) with a functional surface possess obvious advantages and were thus rapidly developed for cancer treatment. Many stimuli-responsive materials, such as nanoparticles, polymers, and inorganic materials, have been applied as caps and gatekeepers to control drug release from MSNs. This review presents an overview of the recent progress in the production of pH-responsive MSNs based on the pH gradient between normal tissues and the tumor microenvironment. Four main categories of gatekeepers can respond to acidic conditions. These categories will be described in detail.

由于各种技术问题使得许多疾病的疫苗研究进展缓慢。要解决疫苗研究中遇到的种种困难,就需要有一个系统的方法去精心设计疫苗。纳米技术可以将各种物理、化学、生物学及医学等技术结合起来优化疫苗效果,其为疫苗的发展带来了曙光。本文中作者概述了纳米佐剂的结构特点、设计思路和作用机制,并联系其研究现状和发展趋势,展望了纳米粒子作为HIV疫苗佐剂的可行性和应用前景。

纳米尺度分子内荧光淬灭效应的研究

本实验将自聚发光分子四苯基乙烯(TPE)和聚集淬灭分子阿霉素(DOX)连接为荧光分子复合物(TPEDOX conjugate,TD),并通过荧光光谱分析和扫描电镜观察结合的方法,对TD纳米聚集体中TPE和DOX的荧光淬灭机制进行了研究。实验发现,在TD分子内,TPE和DOX之间发生了"能量传递接力"的现象,即TPE通过荧光能量共振转移将能量传递给DOX,导致自身荧光发射淬灭;DOX由于TD纳米聚集体的形成,发生了聚集淬灭现象,致使TPE转移的能量和DOX自身的荧光发射均被淬灭,达到一个荧光双淬灭的效果。研究中采用荧光分析与电镜观察结合的方法,阐述了将两种具有相反发光性质的荧光分子在纳米尺度的荧光淬灭机理,为纳米尺度上的分子内荧光淬灭研究提供了理论依据。

Overcoming drug efflux-based multidrug resistance in cancer with nanotechnology被引量:10

Multidrug resistance(MDR),which significantly decreases the efficacy of anticancer drugs and causes tumor recurrence,has been a major challenge in clinical cancer treatment with chemotherapeutic drugs for decades.Several mechanisms of overcoming drug resistance have been postulated.Well known P-glycoprotein(P-gp) and other drug efflux transporters are considered to be critical in pumping anticancer drugs out of cells and causing chemotherapy failure.Innovative theranostic(therapeutic and diagnostic) strategies with nanoparticles are rapidly evolving and are anticipated to offer opportunities to overcome these limits.In this review,we discuss the mechanisms of drug efflux-mediated resistance and the application of multiple nanoparticle-based platforms to overcome chemoresistance and improve therapeutic outcome.
