A Chinese Herbal Formula,Wuzi Yanzong Pill(五子衍宗丸),Improves Spermatogenesis by Modulating the Secretory Function of Sertoli Cells被引量:20

Objective: To evaluate the effects of the Chinese herbal formula Wuzi Yanzong Pill (五子衍宗丸, WYP) on the spermatogenesis and specific secretory functions of Sertoli cells in rat model and to investigate the underlying mechanism. Me^ods: Five groups of male Sprague-Dawley rats including the control group, the model group, the low-dose WYP group, the medium-dose WYP group and the high-dose WYP group (5 in each group) were treated daily with vehicle, multiglycosides of Tripterygium wilfordii Hook f (GTW) either alone (20 mg/kg) or followed by WYP (0.5, 1.0, or 2.0 g/kg daily), respectively for 30 days. Serum levels of follicle-stimulating hormone (FSH), inhibin B (INHB) and testosterone (r) were evaluated using enzyme-linked immunosorbent assay. Androgen- binding protein (ABP) gene expression and transferrin (TF) protein expression in testis tissue specimens of all rats were determined using real-time reverse transcriptase polymerase chain reaction and Western blotting analysis, respectively. Histopathological alterations in the testis were determined using Johnsen's score. Results: The toxicity of GTW towards Sertoli cell secretory functions and spermatogenesis was accompanied by increased serum FSH concentrations and decreased INHB and T concentrations. Upregulated ABP mRNA levels, and decreased TF protein expression and Johnsen's scores were detected in the model group compared with the control group (P〈0.05 or P〈0.01). Oral high-dose WYP administrations to GTW-treated rats effectively alleviated all of the GTW- induced changes in specific secretory functions of Sertoli cells (ABP, INHB and TF). Furthermore, serum T level and Johnsen's score of the testis increased greatly compared with the model group (P〈0.01). Conclusion: WYP has the ability to improve the spermatogenesis, possibly through modulating the secretory proteins expression of Sertoli cells.

目的 观察五子衍宗丸对肾精亏虚证大鼠睾丸支持细胞骨架蛋白表达和生精功能的影响.方法 正常SD雄性大鼠25只,随机分为5组,每组5只.分别为正常组,模型组,五子衍宗丸低、中、高剂量组.正常组早晚均予0.5%羧甲基纤维素钠(50 mg/kg)灌胃;模型组上午给予雷公藤多苷(20 mg/kg)灌胃,下午给予0.5% 羧甲基纤维素钠(50 mg/kg)灌胃;低、中、高五子衍宗丸组大鼠上午予雷公藤多苷(20 mg/kg)灌胃,下午分别予0.5,1.0,2.0 g/kg五子衍宗丸灌胃.30 d后,取睾丸及附睾组织.检测大鼠精子总数、前向运动精子百分率,免疫组化法检测睾丸支持细胞中波形蛋白、α-微管蛋白的表达,TUNEL法检测生精细胞的凋亡.结果 和正常组相比,模型组精子总数、前向运动精子百分率下降,睾丸支持细胞中波形蛋白、α-微管蛋白表达下降,生精细胞凋亡增加;与模型组相比,五子衍宗丸中、高剂量组精子总数、前向运动精子百分率均提高;五子衍宗丸低、中、高剂量组睾丸支持细胞波形蛋白、α-微管蛋白的表达均增加,生精细胞的凋亡率下降.结论 五子衍宗丸可通过调控睾丸支持细胞骨架蛋白的表达,抑制生精细胞的凋亡,改善生精功能.

补肺益肾方对肾精亏虚证大鼠睾丸支持细胞和生精功能的影响被引量:5

目的观察补肺益肾方对肾精亏虚证大鼠睾丸支持细胞和生精功能的影响。方法25只SD大鼠随机分为5组:正常组、模型组、五子衍宗方组、补肺益肾方低剂量组、补肺益肾方高剂量组,每组5只。正常组每天早晚给予5%羧甲基纤维素钠[CMC-Na,100 mg/(kg·d)],其余各组每早给予雷公藤多甙[GTW,20 mg/(kg·d)],模型组每晚给予5%CMC-Na,50 mg/(kg·d),五子衍宗方组每晚给予五子衍宗汤[15.8 g/(kg·d)],补肺益肾方低、高剂量组每晚给予补肺益肾方[6.6 g/(kg·d)、26.6 g/(kg·d)],均灌胃30天。计算脏器指数;采用Western Blot检测睾丸支持细胞波形蛋白(vimentin)、紧密连接蛋白11(claudin-11)和转铁蛋白(TF)表达;ELISA法测定血清抑制素B(INHB)水平;HE染色法和Johnsen评分观察并评价睾丸组织病理学变化。结果与正常组比较,模型组附睾指数、睾丸支持细胞vimentin、claudin-11、TF表达、INHB水平和Johnsen评分下降(P<0.05,P<0.01)。与模型组比较,五子衍宗方组和补肺益肾方高剂量组睾丸支持细胞vimentin、claudin-11、TF表达升高(P<0.05,P<0.01),各给药组INHB水平和Johnsen评分均升高(P<0.01)。与五子衍宗方组比较,补肺益肾方高剂量组睾丸支持细胞vimentin表达、INHB水平和Johnsen评分升高(P<0.05,P<0.01)。结论补肺益肾方高剂量可显著改善肾精亏虚证大鼠睾丸支持细胞和生精功能,其疗效优于五子衍宗方。

Effect of Electroacupuncture on Spermatogenesis in Rats with Oligozoospermia of Insufficiency of Shen(Kidney) Essence Syndrome被引量:1

Objective: To assess the effect of electroacupuncture(EA) on expression of cytoskeletal proteins from Sertoli cells(SCs) and spermatogenesis in rats with oligozoospermia of insufficiency of Shen(Kidney)essence syndrome(OIKES).Methods: Twenty healthy male Sprague-Dawley rats were randomly assigned to four groups using a random number table: control,tripterygium glycosides(TG) treatment,sham and EA groups(n=5 in each group).A rat model of OIKES was established by oral gavage with TG.The EA group was treated with TG and received EA at Shenshu(BL 23) and Zusanli(ST 36) acupoints for 20 min,once daily for 30 days,while the sham group received EA at identical acupoints with skin penetration without stimulation.After 30 days,the ?nal body weight and coef?cients for the testis and epididymis were calculated and sperm parameters were measured.Immunohistochemical analyses were performed to detect expression of vimentin and α-tubulin in SCs and proliferating cell nuclear antigen(PCNA) immunoreactivity in germ cells.Apoptosis in germ cells was quanti?ed by the transferase biotin-dUTP nick end labeling assay.Results: Compared with the control group,the final body weight and testis/epididymis coefficients of rats in the TG-treated group were not significantly different,but the sperm count and motility were lower(P<0.05).Expressions of vimentin and α-tubulin were also signi?cantly weaker(P<0.01).The PCNA immunoreactivity of germ cells was decreased(P=0.059),whereas the apoptotic index of germ cells was increased signi?cantly(P<0.01).In contrast,EA at BL 23 and ST 36 acupoints signi?cantly improved the ?nal body weight as well as the sperm count,concentration and motility(P<0.01 or P<0.05).EA increased expression of vimentin and α-tubulin in SCs markedly,and signi?cantly enhanced PCNA immunoreactivity with decreased apoptosis in germ cells(P<0.01 or P<0.05).Conclusions: EA at BL 23 and ST 36 acupoints has protective effects on spermatogenesis in rats with OIKES.This effect seems to be achieved by attenuating TG-induced disrup
