Moxibustion upregulates hippocampal progranulin expression被引量:6

In China,moxibustion is reported to be useful and has few side effects for chronic fatigue syndrome,but its mechanisms are largely unknown.More recently,the focus has been on the wealth of information supporting stress as a factor in chronic fatigue syndrome,and largely concerns dysregulation in the stress-related hypothalamic-pituitary-adrenal axis.In the present study,we aimed to determine the effect of moxibustion on behavioral symptoms in chronic fatigue syndrome rats and examine possible mechanisms.Rats were subjected to a combination of chronic restraint stress and forced swimming to induce chronic fatigue syndrome.The acupoints Guanyuan(CV4) and Zusanli(ST36,bilateral) were simultaneously administered moxibustion.Untreated chronic fatigue syndrome rats and normal rats were used as controls.Results from the forced swimming test,open field test,tail suspension test,real-time PCR,enzyme-linked immunosorbent assay,and western blot assay showed that moxibustion treatment decreased m RNA expression of corticotropin-releasing hormone in the hypothalamus,and adrenocorticotropic hormone and corticosterone levels in plasma,and markedly increased progranulin m RNA and protein expression in the hippocampus.These findings suggest that moxibustion may relieve the behavioral symptoms of chronic fatigue syndrome,at least in part,by modulating the hypothalamic-pituitary-adrenal axis and upregulating hippocampal progranulin.

温和灸对慢性内脏痛敏大鼠CRF-CRFR1信号途径的调节作用被引量:3

目的:观察温和灸对慢性内脏痛敏大鼠外周(结肠)促肾上腺皮质激素释放因子(CRF)及其受体1(CRFR1)表达的影响。方法:采用乳鼠结直肠扩张(CRD)刺激建立慢性内脏痛敏大鼠模型,选取双侧天枢、上巨虚穴进行温和灸干预。以腹部撤回反射(AWR)评分作为检测大鼠内脏痛敏的行为学指标;采用免疫组织化学法、实时荧光定量PCR法分别观察CRF、CRFR1蛋白及其基因在大鼠结肠的表达及温和灸对其的影响。结果:在不同强度的CRD刺激下,与正常组相比,模型大鼠AWR评分升高(P<0.01)。温和灸组干预后,大鼠AWR评分下降(P<0.01)。模型组大鼠CRF、CRFR1蛋白及mRNA表达均高于正常组(P<0.01),温和灸干预后,CRF、CRFR1蛋白及mRNA表达均降低(P<0.01)。假灸组与模型组比较,差异无统计学意义。结论:温和灸降低大鼠内脏痛敏与其抑制外周(结肠)CRF-CRFR1信号途径有关。
