丹瓜方对ApoE^-/-糖尿病模型小鼠糖脂代谢及血管细胞黏附分子-1及mRNA表达水平的影响被引量:23

目的研究丹瓜方对载脂蛋白E基因敲除(ApoE^(-/-))小鼠复合糖尿病模型糖脂代谢指标、血管细胞黏附分子-1(vascular cell adhesion molecule-1,VCAM-1)及其mRNA表达水平的影响,探讨丹瓜方防治糖尿病合并血管病变的部分机制。方法 Apo E^(-/-)小鼠皮下注射链脲佐菌素(STZ)制备糖尿病模型。将32只成模鼠按照体重分层,再按血糖水平以随机数字表法分为模型组[无菌水(SW),15 m L/(kg·d)]、丹瓜方组[丹瓜方(DG),15 m L/(kg·d)]、联合用药组[吡格列酮(PG),4.3 mg/(kg·d)加DG1 5 mL/(kg·d)]、吡格列酮组[PG,4.3 mg/(kg·d)],每组8只。选血糖正常的8只同龄同品系C 57小鼠作为对照组。灌胃干预12周。监测各组小鼠空腹血糖(FBG)、体重、摄食量及饮水量;测量皮温及鼠尾长度,并通过肉眼观察肝脂肪变程度;测定各组小鼠血清HbA1 c、TC、LDL-C水平,采用放射免疫法测定ET-1,生化法—硝酸还原酶法测定NO,E LISA法测定VCAM-1,应用实时荧光定量PCR(qPCR)检测肾组织VCAM-1 mRNA表达。结果与对照组比较,模型组小鼠体重及摄食量减少,饮水量增加(P<0.05),且血糖值曲线全程高于对照组(P<0.01)。与模型组比较,丹瓜方组小鼠体重增量、HbA1 c、TC、LDL-C、ET-1、VCAM-1水平显著降低,皮温升高(P<0.05或P<0.01)。与吡格列酮组比较,丹瓜方组小鼠体重、体重增量、FBG、TC及LDL-C水平降低(P<0.05或P<0.01)、饮水量和摄食量增加,鼠尾较长(P<0.01);各组小鼠NO水平比较,差异无统计学意义。与对照组比较,模型组肝脂肪变程度较重(P<0.05);与模型组及吡格列酮组比较,丹瓜方组脂肪变程度明显减轻(P<0.05,P<0.01),吡格列酮组肝脂肪变程度重于模型组(P<0.01),而联合用药组肝脂肪变程度处于丹瓜方组和吡格列酮组之间。丹瓜方组VCAM-1 mRNA的表达显著低于模型组(P<0.01),且低于联合用药组和吡格列酮组(P<0.05)。结论丹瓜方对自发性动脉粥样硬化和高脂血症的ApoE^(-/-)

Paradox of Using Intensive Lowering of Blood Glucose in Diabetics and Strategies to Overcome It and Decrease Cardiovascular Risks被引量:6

Hyperglycemia significantly increases the risk of cardiovascular disease(CVD) in diabetics.However,it has been shown by a series of large scale international studies that intensive lowering of blood glucose levels not only has very limited benefits against cardiovascular problems in patients,but may even be harmful to patients at a high risk for CVD and/or poor long-term control of blood glucose levels.Therefore,Western medicine is faced with a paradox.One way to solve this may be administration of Chinese herbal medicines that not only regulate blood glucose,blood fat levels and blood pressure,but also act on multiple targets.These medicines can eliminate cytotoxicity of high glucose through anti-inflammatory and anti-oxidant methods,regulation of cytokines and multiple signaling molecules,and maintenance of cell vitality and the cell cycle,etc.This allows hyperglycemic conditions to exist in a healthy manner,which is called "harmless hyperglycemia" Furthermore,these cardiovascular benefits go beyond lowering blood glucose levels.The mechanisms of action not only avoid cardiovascular injury caused by intensive lowering of blood glucose levels,but also decrease the cardiovascular dangers posed by hyperglycemia.

丹瓜方对GK糖尿病-动脉粥样硬化复合模型大鼠血糖、食量和体重的影响被引量:10

目的:探讨丹瓜方对高脂饲料喂养的GK糖尿病-动脉粥样硬化复合模型大鼠血糖、食量和体重的影响。方法:40只成模GK大鼠,随机分为二甲双胍组、模型对照组、丹瓜方组、辛伐他汀组,自由进食高脂饲料,并予以相应的药物干预;另设10只同龄Wistar大鼠为正常对照组,喂食普通饲料;观察大鼠血糖、HbA1c、体重、食量的变化情况。结果:丹瓜方组血糖与空白对照组比较无差异,且优于其他各模型组;丹瓜方组HbA1c与二甲双胍组比较无差别,且低于模型对照组(P<0.05);GK大鼠食量均明显减少,丹瓜方组食量低于空白对照组和模型对照组(P<0.01);GK大鼠体重均低于空白对照组,丹瓜方组体重增幅最小(P<0.05)。结论:丹瓜方在控制高脂饲料喂养的GK大鼠的血糖、体重和食量上具有明显优势。

Meta-analysis about Western Medicine combined with activating blood drugs on modulating blood glucose and lipids in diabetic patients

OBJECTIVE:To assess the effect of activating blood drugs(AB drugs) on blood glucose and lipid levels of diabetic patients.METHODS:Systematic searches were conducted on 6 electronic databases and the search time was cut off in May 2016.Randomized controlled trials(RCTs) were included if they investigated diabetic patients who received Western medicine combined with AB drugs in the experimental groups and solitary usage of corresponding Western medicine or placebo in the control groups.6 outcome measures,which were considered as inclusion,were changes in fasting blood glucose(FBG),glycosylated haemoglobin A1 c(HbA1c),total triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-c) and high density lipoprotein cholesterol(HDL-c).The literature quality was assessed by Cochrane score for risk of bias in Review Manager 5.2,as well as meta-analysis was performed.RESULTS:7 RCTs comprising 999 patients met all inclusion criteria.Meta-analysis showed beneficial effects of Westernmedicine combined with AB drugs on modulating FBG,TC,LDL-c and HDL-c compared with Western medicine or placebo(P<0.05),while no difference was observed in HbA1c,and TG(P>0.05).To some extent,the therapeutic effects of Western medicine combined with AB drugs were better than that of solitary usage of Western medicine or placebo.Besides,AB drugs were safe and reliable with little toxic and side-effects.CONCLUSIONS:The meta-analysis suggests beneficial effects of Western medicine combined with AB drugs on modulating blood glucose and lipids in diabetic patients.

丹瓜方对HepG2细胞胰岛素抵抗模型LKB1-AMPK-ACC信号通路的影响被引量:2

目的观察丹瓜方对HepG2细胞胰岛素抵抗模型LKB1-AMPK-ACC信号通路的影响,以探讨丹瓜方改善胰岛素抵抗的机制。方法利用高浓度葡萄糖与软脂酸共同体外培养诱导HepG2细胞24 h后成功建立HepG2细胞胰岛素抵抗模型。同时予不同浓度的丹瓜方含药血清干预,以二甲双胍含药血清作为阳性对照。检测培养上清液里残存葡萄糖水平,计算胰岛素敏感指数,检测细胞内的三酰甘油含量,Western blot检测各组LKB1、P-AMPK、AMPK、P-ACC、ACC蛋白表达。结果与模型组相比,二甲双胍组、5%丹瓜方组和10%丹瓜方组的糖耗量、胰岛素敏感指数明显升高(P<0.01),TG含量明显下降(P<0.01),LKB1、P-AMPKα、P-ACC蛋白表达增加(P<0.01),ACC蛋白表达降低(P<0.01)。结论丹瓜方可有效改善高糖高脂诱导的胰岛素抵抗,其分子机制可能与丹瓜方调控LKB1-AMPK-ACC信号通路相关蛋白的表达有关。

丹栝方干预糖尿病动脉粥样硬化大鼠糖脂代谢及氧化应激研究被引量:33

目的探讨丹栝方对糖尿病动脉粥样硬化大鼠糖脂代谢及血清活性氧族(ROS)、胸主动脉核因子κB(nuclear factor kappa B,NF-κB)阳性表达及其mRNA表达水平的影响,以揭示其干预糖尿病慢性并发症的部分机制。方法选取自发性糖尿病GK大鼠(Goto-Kakisaki Wistar rats)40只,以含代谢抑制剂丙硫氧嘧啶的高脂饲料饲养及内皮型一氧化氮合酶(NOS)抑制剂N-硝基-L-精氨酸甲酯腹腔注射,制备高脂糖尿病动脉硬化模型。将GK糖尿病成模鼠先按体重分层,再按血糖浓度根据随机数字表法分为丹栝方组[丹栝方,8mL/(kg·d)]、二甲双胍组[二甲双胍,150mg/(kg·d)]、辛伐他汀组[辛伐他汀,2mg/(kg·d)]、模型组[纯净水,8mL/(kg·d)],每组各10只;另取同龄、体重可比Wistar大鼠10只作为正常对照组(正常组)。各组均灌胃干预24周。监测空腹血糖(FBG)、体重,测定HbA1c、TC、LDL-C、HDL-C、TG、血清ROS,免疫组化法分析主动脉NF-κB,Real-time PCR检测主动脉NF-κB(P65) mRNA表达。结果正常组体重最终显著超过其他各组(P<0.01),四组GK造模组间比较,差异无统计学意义。四组GK造模鼠全程FBG都高于正常组(P<0.01,P<0.05);首次FBG、基线血糖各组间比较,差异无统计学意义(P>0.05),末次FBG二甲双胍组和丹栝方组都显著低于模型组(P<0.01)和辛伐他汀组(P<0.05)。干预24周后,四组GK造模鼠FBG、HbA1c、TC、LDL-C、HDL-C、胸主动脉NF-κB的阳性表达率,及模型组、二甲双胍组、丹栝方组胸主动脉NF-κB mRNA的表达均高于正常组(P<0.01,P<0.05);二甲双胍组、丹栝方组、辛伐他汀组TG、血清ROS,及辛伐他汀组胸主动脉NF-κB mRNA表达低于正常组(P<0.01,P<0.05)。药物干预三组FBG、TC、LDL-C、血清ROS、胸主动脉NF-κB mRNA的表达,丹栝方组、二甲双胍组胸主动脉NF-κB的阳性表达率,及丹栝方组HbA1c、TG显著低于模型组(P<0.01,P<0.05)。二甲双胍组、丹栝方组FBG低于辛伐他汀组(P<0.05)。辛伐�

Dan-gua Fang (丹瓜方) Improves Glycolipid Metabolic Disorders by Promoting Hepatic Adenosine 5'-monophosphate Activated Protein Kinase Expression in Diabetic Goto-kakizaki Rats被引量:12

Objective:To investigate the effect of Dan-gua Fang(丹瓜方) on adenosine 5'-monophosphate(AMP) activated protein kinase(AMPK) α expression in liver and subsequent improvement of glucose and lipid metabolism.Methods:Forty 13-week-old diabetic Goto-Kakizaki(GK) rats were randomly divided into model,Dan-gua Fang,metformin and simvastatin groups(n=10 for each),and fed high-fat diet ad libitum.Ten Wistar rats were used as normal group and fed normal diet.After 24 weeks,liver expression of AMPK α mRNA was assessed by real-time PCR.AMPK α and phospho-AMPK α protein expression in liver was evaluated by Western blot.Liver histomorphology was carried out after hematoxylin-eosin staining,and blood glucose(BG),glycosylated hemoglobin A1c(HbA1c),food intake and body weight recorded.Results:Similar AMPK α mRNA levels were found in the Dan-gua Fang group and normal group,slightly higher than the values obtained for the remaining groups(P<0.05).AMPK α protein expression in the Dan-gua Fang group animals was similar to other diabetic rats,whereas phospho-AMPK α(Thr-172) protein levels were markedly higher than in the metformin group and simvastatin group(P<0.05),respectively.However,phosphor-AMPKa/AMPK α ratios were similar in all groups.Dan-gua Fang reduced fasting blood glucose with similar strength to metformin,and was superior in reducing cholesterol,triglycerides,high-density lipoprotein cholesterol as well as improving low-density lipoprotein cholesterol in comparison with simvastatin and metformin.Dan-gua Fang decreases plasma alanine aminotransferase(ALT) significantly.Conclusion:Dan-gua Fang,while treating phlegm-stasis,could decrease BG and lipid in type 2 diabetic GK rats fed with high-fat diet,and effectively protect liver histomorphology and function.This may be partly explained by increased AMPK expression in liver.Therefore,Dan-gua Fang might be an ideal drug for comprehensive Intervention for glucose and lipid metabolism disorders in type 2 diabetes mellitus.

目的探讨丹瓜方对Apo E^-/-糖尿病模型小鼠脑组织胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)表达水平的影响,以探究丹瓜方防治糖尿病脑病的部分机制。方法以链脲佐菌素(STZ)对Apo E^-/-小鼠塑造糖尿病模型。将成模后的小鼠按体质量分层,依照随机数字表随机分为模型组、丹瓜方组、吡格列酮组、联合组,每组8只。选取血糖正常的同龄同品系C57小鼠8只作为对照组。分组灌胃干预12周。ELISA法测定IGF-1表达。结果与C57组IGF-1(19.89±0.95)ng/m L比较,其余各组均明显降低(P<0.01);丹瓜方组(14.31±0.99)ng/m L和联合用药组IGF-1水平(11.58±0.92)ng/m L明显高于模型组(7.25±0.42)ng/m L;丹瓜方组IGF-1水平明显高于联合用药组(P<0.01)。结论通过适度提升糖尿病鼠脑组织中IGF-1表达,可能是丹瓜方防治糖尿病脑病的部分机制。
