棉纤维/季铵盐接枝共聚物的合成及抗菌性能研究被引量:7

采用硝酸铈铵为引发剂,在棉纤维表面引发甲基丙烯酸二甲氨基乙酯(DMAEMA)接枝聚合,并使接枝的DMAEMA季铵化,以赋予棉纤维良好的抗菌性能。实验对影响DMAEMA接枝率的关键因素进行了细致研究,发现,当φ(DMAEMA)为10%,引发剂浓度为20 mmol/L,pH为1~1.5时,在45℃反应5 h后,得到较好的接枝效果。同时发现,溶剂类型对棉纤维/DMAEMA接枝共聚物的季铵化反应影响很大,丙酮对季铵化最为有利。接枝率的提高可加强棉纤维/季铵盐接枝共聚物的抗菌性能;与单独采用溴代正丁烷为季铵化试剂相比,先后采用溴代正丁烷和溴代十二烷作为季铵化试剂,所得材料的抗菌性能更为优良,说明长链季铵盐聚合物与短链季铵盐聚合物间存在协同抗菌作用。

Preparation and Antibacterial Activity of Copolymer of Methacryloxyethyldimethyl Dodecyl Ammonium Bromide/Acrylamide

Copolymers PMDAB-co-AA with excellent antibacterial activity were designed and prepared(MDAB/AA=methacryloxyethyldimethyl dodecyl ammonium bromide with acrylamide).The chemical structures of the copolymers were characterized by Fourier transform infrared(FTIR),1 H NMR,13 C NMR and chemical titration.The antimicrobial activities and kinetics of the copolymer against E.coli and S.aureus were examined by viable cell counting method.The compositions of the copolymers could be easily controlled by the feed molar ratio of MDAB to AA.Furthermore,the highest antibacterial activity was achieved when the molar fraction of MDAB was in a range of 5%―20%,and the killing rate reached 100% under the test conditions.In addition,the antibacterial activity of PMDAB-co-AA was maintained and stable without any loss after 15 times of repeated usage.It was proved that the PMDAB-co-AA samples targeted at cell membrane,and caused serious damage to cell integrity and inner membrane permeation.The surrounding conditions,such as pH and inorgainc salts concentrations(CaCl2 or NaCl),also affected their antibacterial activities.

比较了高分子抗菌剂和小分子抗菌剂在作用方式上的区别,说明了高分子抗菌剂具有抗菌活性和选择性强、效果持久、安全无毒等优点,具有良好的研究价值。还介绍了高分子抗菌剂的研究现状,包括主要类型、结构特点、抗菌机理、构效关系等方面的内容,旨在为后续研究提供参考。最后对高分子抗菌剂的应用前景作了展望,认为其有望在水净化、医疗设备和卫生用品等对安全性要求更高的领域将获得大量应用。

Synthesis and Antibacterial Action of Poly(DMAEMA-BC) with Various Molecular Weights

Antibacterial polymers of dimethylaminoethyl methacrylate benzyl ammonium chloride(DMAEMA-BC) with various molecular weights(Mws) were prepared under controlling radical polymerization conditions. The Mws of these polymers were determined by means of static multiangle laser light scattering and viscosity method. A Mark-Houwink equation was established to be [η]=0.154M0.764 for the Mw evaluation of poly(DMAEMA-BC)s. The effects of the Mws of these poly(DMAEMA-BC)s on their antibacterial activities against E. coli and S. aureus were investigated by various methods including viable cell counting, electrical conductivity titration, intracellular constituent leakage tests and electron microscopy. Our results indicate that the antibacterial efficiency of DMAEMA-BC was significantly enhanced after the monomers were polymerized into a polymer and increased obviously with the Mws,as a result of the increase of charge density. Further investigation of the molecular basis underlying the anti-bacterial role of these polymers revealed that poly(DMAEMA-BC) promoted the release of potassium ion from the membrane of bacterial cells and the release increased significantly with the Mws of the polymers used.

季铵盐类聚合物(PQACs)是高分子抗菌剂的典型代表。本文介绍了PQACs的两种较为普遍认可的抗菌机理,即"穿透型机理"和"离子交换机理",指出PQACs有良好的结构稳定性,能重复持久地抗菌,归纳了分子量、取代烷基链长度、抗衡阴离子、间隔长度这四种结构因素对PQACs与细菌间的静电和疏水作用的影响,建立其构效关系。最后展望了PQACs的发展前景,认为应加强其构效关系和应用技术的研究。
