比色法测定楤木总苷胶囊中的总皂苷被引量:2

目的建立测定楤木总苷胶囊中总皂苷含量的方法。方法采用比色法,以齐墩果酸为对照品,香草醛冰醋酸-高氯酸为显色剂,在554nm处测定,以量比系数2.0进行校正。结果楤木总苷胶囊中总皂苷的含量为10.04%,以每粒胶囊0.25g计含橡木总皂苷25.1mg,线性范围为5.0—22.5μg·mL-1,平均加样回收率为100.12%(RSD=1.19%)。结论所用方法操作简便、快速准确、灵敏度高、重复性好,可用于楤木总苷胶囊的含量测定及质量控制。

楤木皂苷胶囊成型工艺辅料筛选被引量:3

目的:优选楤木皂苷胶囊的成型工艺辅料。方法:以颗粒的成型合格率、吸湿百分率、休止角和堆密度为评价指标,在逐一单个筛选辅料的基础上,筛选出适合制备楤木皂苷胶囊的混合辅料,并考察临界相对湿度(CRH)。结果:微晶纤维素-乳糖(1:1,m/m)混合辅料制成的颗粒抗潮性能强,流动性好。CRH为78%。结论:所选辅料可用于制备楤木皂苷胶囊。

太白楤木总皂苷提取物的质量标准研究被引量:4

目的:建立太白楤木根皮中总皂苷含量的测定方法。方法:采用TLC薄层色谱法,以甲醇-氯仿-水(6.5∶3.5∶1)为展开剂展开后,用10%硫酸乙醇溶液显色,并在105°C下加热进行鉴别。接着以香草醛-冰醋酸法显色,以屏边三七苷为对照品,在检测波长553 nm处对太白楤木总皂苷采用紫外-可见分光光度法进行含量测定。结果:在TLC色谱中,供试品与对照品色谱相应的位置上,日光下显相同的紫色斑点;紫外灯(365 nm)下显相同的紫色荧光斑点。屏边三七苷在0.020 65~0.123 9 g.L-1线性关系良好(r=0.999 9),平均加样回收率为100.20%,RSD 0.88%(n=6),太白楤木根皮中总皂苷中总皂苷含量为61.30%。结论:以太白楤木根皮总皂苷含量为主要评价指标,所建立的方法准确、简便、快速,重复性好,可作为太白楤木根皮中总皂苷含量控制方法。

HPLC法测定楤木皂苷胶囊中齐墩果酸含量被引量:1

目的:建立楤木皂苷胶囊中齐墩果酸的含量测定方法。方法:采用Diamonsil C18(2)(250mm×4.6mm,5μm),甲醇-0.1%磷酸水(80:20)为流动相,检测波长210nm;流速1mL/min,柱温为室温,进样量20μl。结果:齐墩果酸回归方程:Y=9132.4 X-47376,r=0.999 7,在27.5~110μg范围内线性关系良好,平均回收率为100.49%,RSD为0.60%。结论:本方法简便快捷,准确可靠,可用于测定楤木皂苷胶囊中齐墩果酸的含量。

Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro被引量:21

OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. The protection of MLB may contribute to its antioxidant properties.
