一种不依赖于无菌培养的拟南芥活体转基因种子筛选方法被引量:5

拟南芥活体转基因种子的筛选仍主要采用无菌培养技术,存在操作麻烦,容易污染而造成转基因材料损失等问题.本研究在传统组织培养筛选技术的基础上,通过去除原培养基成分中对种子发芽和筛选不必需的蔗糖和有机成分,降低大量元素的浓度,缩短筛选时间,从而免除了传统转基因种子筛选中培养基和种子消毒的繁琐步骤,建立了一种不依赖于无菌培养的转基因种子筛选方法.与无菌培养筛选比较,筛选效果相同,但简化了试验步骤,提高了筛选操作的可靠性.

Codon Usage Biases of Transposable Elements and Host Nuclear Genes in Arabidopsis thaliana and Oryza sativa被引量:6

Transposable elements (TEs) are mobile genetic entities ubiquitously distributed in nearly all genomes. High frequency of codons ending in A/T in TEs has been previously observed in some species. In this study, the biases in nucleotide composition and codon usage of TE transposases and host nuclear genes were investigated in the AT-rich genome of Arabidopsis thaliana and the GC-rich genome of Oryza sativa. Codons ending in A/T are more frequently used by TEs compared with their host nuclear genes. A remarkable positive correlation between highly expressed nuclear genes and C/G-ending codons were detected in O. sativa (r=0.944 and 0.839, respectively, P〈0.0001) but not in A. thaliana, indicating a close association between the GC content and gene expression level in monocot species. In both species, TE codon usage biases are similar to that of weakly expressed genes. The expression and activity of TEs may be strictly controlled in plant genomes. Mutation bias and selection pressure have simultaneously acted on the TE evolution in A. thaliana and O. sativa. The consistently observed biases of nucleotide composition and codon usage of TEs may also provide a useful clue to accurately detect TE sequences in different species.

粳稻光敏型核不育系对日长敏感性的鉴定分析被引量:2

提出利用自然日长鉴定粳稻光敏不育系的方法。应用AMMI(additive main effects and multiplicative interaction)模型,对8个水稻光敏不育系对日长和温度反应的遗传敏感性进行比较分析,将其育性敏感期日长分成6个梯度,温度分成11个梯度。结果表明:在杭州自然条件下种植参试的8个不育系均为光敏核不育系,具有以下特性,抽穗前10~20d是育性转化敏感期,不育系的可育临界日长为13.0~13.8h,育性的临界高温为25.0℃,临界低温为21.0℃。

Fine mapping and candidate gene prediction of the photoperiod and thermo-sensitive genic male sterile gene pms1(t) in rice被引量:10

Pei'ai64S, an indica sterile variety with photoperiod and thermo-sensitive genic male sterile (PTGMS) genes, has been widely exploited for commercial seed production for "two-line" hybrid rice in China. One PTGMS gene from Pei'ai64S, pmsl(t), was mapped by a strategy of bulked-extreme and recessive-class approach with simple sequence repeat (SSR) and insert and deletion (In-Del) markers. Using linkage analysis for the F2 mapping population consisting of 320 completely male sterile individuals derived from a cross between Pei'ai64S and 93-11 (indica restorer) lines, the pmsl(t) gene was delimited to the region between the RM21242 (0.2 cM) and YF11 (0.2 cM) markers on the short arm of chromosome 7. The interval containing the pmsl(t) locus, which was co-segregated with RM6776, is a 101.1 kb region based on the Nipponbare rice genome. Fourteen predicted loci were found in this region by the Institute for Genomic Research (TIGR) Genomic Annotation. Based on the function of the locus LOC_Os07g12130 by bioinformatics analysis, it is predicted to encode a protein containing a Myb-like DNA-binding domain, and may process the transcript with thermosensory response. The reverse transcription-polymerase chain reaction (RT-PCR) results revealed that the mRNA levels of LOC_Os07g12130 were altered in different photoperiod and temperature treatments. Thus, the LOC_Os07g12130 locus is the most likely candidate gene for pmsl(t). These results may facilitate not only using the molecular marker assisted selection of PTGMS genes, but also cloning of the pmsl(t) gene itself.

分离缓冲液对水稻细胞核悬液DNA分辨率效应的比较被引量:19

为制备高纯度水稻细胞核悬液供FACSCalibur流式细胞仪使用,选用6种细胞核分离缓冲液制备样品,对其DNA分辨率效果进行比较,Otto buffers和Marie’s nuclearisolation buffer表现分辨率较高,细胞G0/1峰产生的变异系数较低(CV分别为3.57%和3.41%);其中Otto buffers的组分较Marie’s nuclear isolation buffer简单、成本低,且能在正常室温条件下分析和保存。虽然在细胞核悬液中Tris-MgCl2buffer显示最低的CV值(3.19%),但产生较多的背景碎片。而其余3种缓冲液(Lysis buffer LB01,Galbraith’s buffer和Arumuganathan’sbuffer)的变异系数均较高(分别为4.75%,6.28%和6.73%)。结果表明采用Otto buffers分离缓冲液制备细胞核悬浮液,能实现苗期对水稻花粉植株的不同倍性(单倍体、二倍体、三倍体等)快速鉴定。

不同倍性水稻植株茎解剖结构比较研究被引量:7

为研究倍性变化对水稻植株茎结构的影响,以粳稻FC311和FC322为供试品系,应用流式细胞术从花培后代中鉴定出6份不同倍性的材料(FC311中的单倍体、二倍体及三倍体;FC322中的单倍体、二倍体及四倍体),利用石蜡切片法观察并比较不同倍性植株茎的解剖结构.结果表明:供试材料节间维管束数及大小、茎壁面积、维管束面积通常与倍性的增加有关,显示明显的基因剂量效应;而表皮下和内、外圈维管束鞘的厚壁细胞层数、发育特征及细胞淀粉粒含量等结构和特性,与倍性变化无明显相关;在FC322四倍体植株第1节间中,还观察到颇为少见的2层厚壁组织结构.

台风对水稻温敏核不育系育性的影响被引量:1

2004年度登陆浙江的4个台风"云娜"(RANANIM)、"艾利"(AERE)、"桑达"(SONGDA)、"海马"(HAIMA)产生不同程度的降温效应,"桑达"和"海马"导致杭州日平均最低温度低于25℃,并持续数天,对水稻温敏不育系育性产生了明显影响。台风侵扰期间在杭州自然条件下观察鉴定了11个温敏不育系花粉育性的转换特性。在持续<25℃的低温2d下,不育系育性并无影响,但日平均温度和日最低温度<25℃持续4d以上时,则会导致不育系育性转换。台风到达后10~12d,多数温敏不育系会出现正常花粉比率高峰,将对两系法杂交水稻种子生产有严重影响。

细胞质雄性不育和恢复系统(CMS/Rf)在植物杂种优势利用中已被广泛应用。为阐明恢复基因在这一系统中的作用机理,众多研究者开展了恢复基因的定位和克隆研究。近年来,4个植物恢复基因的成功克隆有力地推动了这一研究领域的发展。文章综述了植物恢复基因的定位、克隆以及育性恢复分子机理的研究进展,并讨论了恢复基因在植物分子育种上的应用。
