侧链烯丙酯化的生物可降解聚碳酸酯的酶促合成与功能化被引量:1

报道了多孔硅球固定化猪胰脂肪酶(IE)催化合成侧链含活性烯丙氧羰基的聚(5-甲基-5-烯丙氧羰基-三亚甲基碳酸酯)(PMAC)的新方法.所得聚合物经核磁共振氢谱(1H NMR),核磁共振碳谱(13C NMR)以及凝胶渗透色谱(GPC)表征.同时,研究了反应温度及反应时间对聚合反应的影响.结果表明:严格控制反应温度75℃、反应时间120 min,可得到线型的PMAC分子,其数均分子量(Mn)和重均分子量(Mw)分别达到34 700和53 800,分散度为1.55.进一步研究了所得聚合物的功能化,即侧链环氧化,12 h可实现其完全的环氧化.

Synthesis and Characterization of Novel Aliphatic Polycarbonates Bearing Pendant Allyl Ether Groups

A new six-membered cyclic carbonate monomer, 5-allyloxytrimethylene carbonate (ATMC), was synthesized starting from glycerol, and the corresponding polycarbonates, poly(5-allyloxytrimethylene carbonate)(PATMC) were further synthesized by ring-opening polymerization in bulk at 150℃ using stannous octanoate as an initiator. The structures of the monomer and the polymers were confirmed by IR, IH-NMR, 13C-NMR, and GPC analysis.

Enzymatic Synthesis and Characterization of Novel Amphiphilic Triblock Copolymer Poly(p-dioxanone-co-5-benzyloxytrimethylene carbonate)-block-poly(ethylene glycol)被引量:1

Novel amphiphilic triblock copolymer poly(p-dioxanone-co-5-benzyloxytrimethylene carbonate)-block-poly(ethylene glycol)-block-poly(p-dioxanone-co-5-benzyloxytrimethylene carbonate) (p(PDO-co-BTMC)-b-PEG-b-p(PDO-co-BTMC)) was successfully synthesized using immobilized porcine pancreas lipase on porous silica particles (IPPL) as the catalyst for the fLrSt time. 1H NMR, 13C NMR and GPC analysis were used to confirm the structures of resulting copolymers. The molecular weight (Mn) of the copolymer with feed ratio of 69:20:11 (BTMC: PDO: PEG ) was 31300 g/mol and the polydispersity was 1.85, while the Mn decreased to 25000 g/mol and polydispersity of 1.93 with the feed ratio of 50:40:10.

报道了多孔硅球固定化猪胰脂肪酶(IE)催化一步合成侧链含活性羧基的聚(酯 碳酸酯)的方法.所得共聚物经红外光谱(IR)、核磁共振氢谱(1HNMR)和特性粘度表征.研究了反应温度、反应时间、IE用量及共聚单体5 甲基 5 羧基三亚甲基碳酸酯(MCC)和丙交酯(DLLA)的摩尔投料比n(MCC)/n(DLLA)对共聚反应的影响.结果表明:反应温度150℃,IE用量0.5%,反应时间48h,n(MCC)/n(DLLA)=1∶4为最佳的共聚反应条件.在此条件下,聚合物的粘度和产率分别达到9.86mL·g-1和89.6%.

Synthesis and Characterization of Amphiphific Block Copolymer Containing PVP and Poly(5-benzyloxytrimethylene carbonate)被引量:5

Amphiphilic copolymer of 5-benzyloxytrimethylene carbonate (BTMC) with poly (vinyl pyrrolidone) (PVP) was successfully synthesized using immobilized porcine pancreas lipase (IPPL) or SnOct2 as catalyst. Hydroxyl terminated PVP, synthesized with 2-mercaptoethanol as a chain transfer reagent, was employed as a rnacroinitiator. The resulting copolymers were characterized by GPC, ^1H NMR and IR. Increasing the BTMC/PVP-OH feed ratio ([B]/[P]) resulted in the increase of Mn of corresponding copolymers and the decrease of Mw/Mn. Immobilized enzyme has comparable catalytic activity to SnOct2 for the copolymerization.

Enzymatic Synthesis and Characterization of Novel Polycarbonates Derived from 5-Allyloxy-1, 3-dioxan-2-one and 5, 5-Dimethvl-1.3-dioxan-2-one

Novel polycarbonates from 5-allyloxy-1, 3-dioxan-2-one (ATMC) with 5, 5-dimethyl- 1, 3-dioxan-2-one (DTC) were successfully synthesized for the first time using immobilized porcine pancreas lipase (IPPL) as catalyst. The resulting copolymers were characterized by IR, ^1H NMR, ^13C NMR and GPC. The molecular weight (Mn) of the copolymer with molar feed ratio of 10:90 (ATMC:DTC) was 9300 and the polydispersity was 1.31, while the Mn increased to 14300 and polydispersity of 1.25 with the feed ratio of 50:50. Moreover, the composition of the copolymers agreed well with the monomer feed.

Porcine pancreas lipase(PPL) immobilized on narrow distributed micron glass beads was employed successfully for ring-opening polymerization of ε-caprolactone(CL).Different polymerization conditions such as enzyme concentration,reaction temperature and reaction time were studied.The results showed that M_n of the resulting PCL was significantly increased compared with that catalyzed by PPL.Higher temperature and longer reaction time both contributed to gaining PCL with a higher molecular weight,while the yield had almost no change.In addition,for evaluating the recyclibilty of immobilized PPL on narrow distributed micron glass beads for the ring-opening polymerization of CL,we adopted the most severe reaction conditions(180 ℃,(240 h)) in the recycling experiments.It was found that the recovered immobilized PPL could be used again with a compatible high catalytic activity.The highest M_n of 21 300 of PCL could be obtained at a mass fraction of 5.18% of the reused immobilized PPL at 180 ℃ for 240 h,which is the highest M_n of PCL catalyzed by PPL.The excellent recyclability of immobilized PPL on narrow distributed micron glass beads is very helpful to its further industry applications.

SYNTHESIS OF POLY(5,5-DIMETHYL-1,3-DIOXAN-2-ONE)BY LIPASE-CATALYZED RING-OPENING POLYMERIZATION被引量:1

Porcine pancreas lipase (PPL) and PPL immobilized on narrow distributed micron-sized glass beads wereemployed successfully for the ring-opening polymerization of 5, 5-dimethyl-1, 3-dioxan-2-one (DTC) for the first time.Different polymerization conditions such as enzyme concentration and reaction temperature were studied. Immobilized PPLexhibits higher activity than native PPL. Along wth the increasing enzyme concentration, the molecular weigh of resultingPDTC decreases. PPL immobilized on narrow distributed micron-sized glass beads has outstanding recyclability. For thethird recycle time, immobilized PPL exhibits the highest catalytic activity and with high activity even after the fifth recyletime for the synthesis of PDTC. The ~1H-NMR spectra indicate that decarboxylation does not occur during the ring-openingpolymerization.

研究了5,5 二甲基 三亚甲基碳酸酯的均聚物(PDTC)及其与聚乙二醇(PEG)组成的嵌段共聚物(PDTC PEG PDTC)的亲水性、水解和酶解性能以及药物释放性能.结果表明:随着聚合物中PEG含量的增加,聚合物的亲水性显著增强;当PEG含量较高时,PEG从聚合物链上断裂溶解使得聚合物的降解速率相对较快;在猪胰脂肪酶(PPL)或假丝酵母皱褶酶(CL)的催化下水解108h,PEG含量对聚合物的水解速率影响不大;药物释放速率随着PEG含量的增大明显提高,n(EO)/n(DTC)为4∶1的嵌段共聚物在30min时释药率高达95%.

以猪胰脂肪酶或假丝酵母皱褶酶为催化剂,100℃下通过本体聚合成功地合成了三亚甲基碳酸酯(TMC)和2-的乙氧基-2-氧.1,3-二氧磷杂环戊烷(EEP)的无规共聚物(poly(TMC-co-EEP).研究了酶浓度,聚合反应时间以及共聚单体投料比等因素对共聚物分子量和产率的影响.随着酶(PPL或CL)浓度的增加,共聚物分子量降低.同时,随着EEP投料比的增加,共聚物的分子量也降低.共聚物数均分子量最大可达到10200.随着共聚单体投料摩尔比(EEP/TMC)从0增加到5:10,共聚物的玻璃化温度从-28℃降低到-41.7℃.体外降解实验表明:共聚物中磷酸酯含量越高,降解速率越快.
