Applications of Three DNA Barcodes in Assorting Intertidal Red Macroalgal Flora in Qingdao, China被引量:7

This study is part of the endeavor to construct a comprehensive DNA barcoding database for common seaweeds in China. Identifications of red seaweeds, which have simple morphology and anatomy, are sometimes difficult solely depending on morphological characteristics. In recent years, DNA barcode technique has become a more and more effective tool to help solve some of the taxonomic difficulties. Some DNA markers such as COI (cytochrome oxidase subunitⅠ) are proposed as standardized DNA barcodes for all seaweed species. In this study, COI, UPA (universal plastid amplicon, domain V of 23S rRNA), and ITS (nuclear internal transcribed spacer) were employed to analyze common species of intertidal red seaweeds in Qingdao (119.3 -121 E, 35.35 -37.09 N). The applicability of using one or a few combined barcodes to identify red seaweed species was tested. The results indicated that COI is a sensitive marker at species level. However, not all the tested species gave PCR amplification products due to lack of the universal primers. The second barcode UPA had effective universal primers but needed to be tested for the effectiveness of resolving closely related species. More than one ITS sequence types were found in some species in this investigation, which might lead to confusion in further analysis. Therefore ITS sequence is not recommended as a universal barcode for seaweeds identification.

我国裙带菜育苗和育种技术的回顾和展望被引量:6

裙带菜(Undaria pinnatifida)是我国最重要的经济海藻之一,其产量和经济价值在褐藻中仅次于海带。我国裙带菜的大规模人工栽培始于20世纪80年代,但是育苗的基础研究早在60年代就已开始。育苗和育种技术对于任何一种大型经济海藻的栽培产业发展都具有至关重要的作用,裙带菜也不例外。近年来,我国的裙带菜育苗和育种技术有了长足的发展,但是仍有很多问题亟待解决。对我国裙带菜育苗和育种技术的发展历史进行扼要回顾,分析其现状和存在的问题,并对其未来的发展趋势进行展望和思考,以期为裙带菜栽培产业未来的发展提供参考。

Spermatozoid life-span of two brown seaweeds, Saccharina japonica and Undaria pinnatifida, as measured by fertilization efficiency被引量:1

During sexual reproduction of seaweeds, spermatozoid (sperm) discharge is triggered by chemical messengers (pheromones) released by the female gametes. The chemotactic ability of the sperm ensures fertilization success. Using unialgal male and female gametophyte material under designated standard gametogenesis testing (SGT) conditions, the potential life-span of the sperm of two seaweeds, Saccharina japonica and Undaria pinnatifida, was assessed by their ability to fertilize eggs. Results show that within 20-30 min after being discharged, sperm of both species could complete fertilization without an apparent decline in fertilization rate. Although fertilization rate 60-120 min after sperm discharge dropped significantly in both species, some sperm were viable enough to fertilize the eggs. In S.japonica, at 12°C, some sperm were able to fertilize eggs up to 12 h after discharge. In both species, egg discharge rates (EDR) in the male and female mixed positive controls were significantly higher than those of all the sperm-testing groups. Doubling the seeded male gametophytes of S. japonica in the SGT tests significantly increased the EDR, further confirming the effect of the presence of the male on the female in terms of facilitating egg discharge from oogonia.

杂交海带新品系B013孢子囊形成规律初探被引量:2

为了了解海带受精时间和孢子囊形成的关系,以B013品系为材料,在2014-2015年度,先后在5月、6月、7月和8月,四次完成受精,培育了B013幼苗,并在大连旅顺海带栽培区开展了栽培试验。结果表明,5、6月份培育的藻体,分别有100%和80%的藻体在1月中旬开始在顶端1/3处形成首批宽度6 cm左右,长度50~60 cm左右的孢子囊群,这些孢子囊在随后的3个月中,逐渐释放了孢子,并随着藻体的不断生长,孢子囊形成区域的藻体逐渐褪去,到了9月中、下旬,藻体全部区域形成第二批孢子囊群;7、8月份培育的藻体没有在冬季形成首批孢子囊,藻体持续生长,只在9月中、下旬,在藻体的全部,形成了唯一的一次孢子囊群。所有从首批、次批孢子囊释放出来的游孢子经过附着、后期培育测试,都能够正常的形成健康的幼孢子体。这些结果表明,B013孢子囊形成规律不同于双亲中的栽培种群父本,而更加像野生的母本,总是在藻体生长到一定程度时候,在顶端先少量形成孢子囊群,之后在秋天日照时间变短,海水温度开始下降的时候大批量形成孢子囊群,藻体随后流失。该研究结果为后续研究B013孢子囊的形成机制提供了一定的理论依据。

DNA条码技术在大型海藻系统学中的研究现状被引量:2

大型海藻(Macroalgae)依据光合色素的不同而分为红藻(Rhodophyta),褐藻(Phaeophyta)和绿藻(Chlorophyta),它们是海洋环境中重要的初级生产者。大型海藻可以直接为人类提供食物,也可以作为饲料转化为动物性蛋白。同时,大型海藻还是重要的药源生物。近年,我国科学家仅利用海带就成功得开发出了PSS、肾海康和甘糖酯等多种药物。此外,由于大型海藻是自养生物,对改善近岸富营养化水域有重要作用。

几种江蓠属海藻3个分子序列的系统学分析被引量:1

分析了我国沿海几种常见的江蓠属(Gracilaria)海藻的18S rRNA基因、cox2-3间隔区以及RUBISCO间隔区的分子序列,并结合GenBank现有的相关数据进行了分子系统学关系分析,为江蓠属的系统进化和分类地位提供了新的佐证。结果表明,基于cox2-3间隔区、以及RUBISCO间隔区序列构建的MP(Maximum parsimony)进化树较为相似,而与基于18S rRNA构建的进化树略有不同。这主要是由于18S rRNA更为保守的原因;扁江蓠与脆江蓠在3个系统树中均聚合成支,显示了它们之间具有较近的亲缘关系;龙须菜与江蓠属海藻具有较远的遗传距离,在3个进化树中,龙须菜也均位于进化树的基部,单独成支,证实龙须菜并不隶属于江蓠属,且分化相对较早。

Intraspecific genetic analysis,gamete release performance,and growth of Sargassum muticum(Fucales,Phaeophyta) from China被引量:2

Sargassum muticum is one of the most abundant and conspicuous native macroalgae species on the northern coasts of China.It often forms large-scale seaweed beds in subtidal zones.This investigation was designed to understand the intraspecific genetic relationships of this alga based on samples collected from four northern coastal sites of China,and to evaluate gamete release and growth capacity in laboratory conditions.The nuclear internal transcribed spacer 2(ITS2) sequences of 16 samples from four locations were identical.Based on cox3 gene and partial rbcLS operon sequences,intraspecific genetic variability was detected with three and two ribotypes,respectively.Temperature,not irradiance,was shown to significantly affect gamete release and fertilization.Elevated temperature and irradiance enhanced the growth of germlings and vegetative branchlets.Maximum growth rate of germlings was detected at 18–24°C and an irradiance of 60–100 μmol photons/(m2 s).Under ambient conditions(12–25°C and 60–125 μmol photons/(m2 s)),relative growth rate of young branchlets could reach 7.5%/d.
