Objective: To investigate the expression of Fas ligand in human colon carcinoma cell lines. Methods: A total of six human colon cancer cell lines were examined for the expression of Fas ligand mRNA and cell surface protein by using RT-PCR and flow cytometry respectively. Results: The results showed that Fas ligand mRNA was expressed in all of the six cancer cell lines and Fas ligand cell surface protein was expressed in part of them. Conclusion: These data suggest that Fas ligand was expressed, at least in part, in human colon cancer cell lines and might facilitate to escape from immune surveillance of the host.
目的 :探讨 c FL IP(细胞型 Fas相关死亡区域蛋白样白介素 - 1β转换酶抑制蛋白 ) m RNA在大肠癌细胞中的表达及其与 Fas抗原 (CD95 )表达的相关性。方法 :应用 RT- PCR方法检测 3株人大肠癌细胞株中 c- FL IP m RNA的表达 ,采用间接免疫荧光标记流式细胞术检测大肠癌细胞 Fas抗原表达。结果 :SW1116及 SW6 2 0细胞株 c- FL IP m RNA表达阳性并相对高水平表达 Fas抗原 ,而 L o Vo细胞株 c- FL IP m RNA表达阴性并低水平表达 Fas抗原。结论 :在大肠癌细胞中 ,c- FL IP m RNA的表达趋势与 Fas抗原一致 ,提示 c- FL IP可能在抑制 Fas抗原诱导的细胞凋亡中发挥重要作用。
