Preparation of luminescent CdTe quantum dots doped core-shell nanoparticles and their application in cell recognition被引量:7

Based on the reverse microemulsion technique, luminescent quantum dots doped core-shell nanoparticles have been prepared by employing silica as the shell and CdTe quantum dots as the core of the nanoparticles, which have an excellent solubility and dispersibility, especially amine and phosphonate groups have been modified on their surface synchronously. In comparison with CdTe quantum dots, these nanoparticles show superiority in chemical and photo-chemical stability. The quantum dots doped core-shell nanoparticles were successfully linked with lactobionic acid by amine group on it, which was used to recognize living liver cells.

利用反向微乳液技术,以CdTe量子点为核,SiO2为壳,一步制备了表面带有氨基和磷酸基团的核壳型量子点荧光纳米颗粒.该颗粒水溶性好,大小均匀,有效改善了CdTe量子点的不稳定性,抗光漂白性能大大增强.结合纳米技术、生物技术与荧光标记技术,基于硅壳包被的核壳型量子点荧光纳米颗粒,成功实现了对肝实质细胞的识别.

分子信标用于p53 mRNA的体外定量检测被引量:9

根据p53基因的序列设计并合成了能特异性检测p53mRNA的分子信标(MB),发展了一种快速定量测定细胞内总RNA提取物中p53mRNA的方法.采用鼻咽癌(CNE2)细胞系和经RNA干扰技术降低p53基因表达的CNE2-p53RNAi细胞系,抽提总RNA并用MB检测,验证了MB的检测对象是p53mRNA.将该方法应用于多种肿瘤细胞内p53基因表达水平的分析,表达变化趋势与经典的mRNA分析方法RT-PCR检测结果相符.在此基础上,用MB对5-氟尿嘧啶(5-Fu)处理的肺腺癌细胞(A549)进行了p53mRNA的体外定量检测,结果表明采用MB能够快速地获知该药物对细胞内p53mRNA表达影响的信息.

In order to conduct the fundamental investigation of the effect of 5-FU on HNE1 cells,p33ING1 mRNA level in cellular total RNA was detected quantitatively based on MB assay.During the operation,the effects of 5-FU concentrations and treatment time in HNE1 cells and HNE1 cells transfected with p33ING1 were measured in vitro.The results were as follows: p33ING1 mRNA expression level in tumor cells was enhanced not only by the 5-FU concentration but also by lapse of time.The MTT results also proved that high expression of p33ING1 mRNA can increase cell’s sensitivity to chemical drug 5-FU.The detection method based on MB can be used to provide useful evidence quickly and quantitatively for gene expression and new chemical drugs development.
