Protective effect of Angelica sinensis on cerebral neurons from rat embryos under hypoxia被引量:7

BACKGROUND: The enhanced expression of c-Fos protein in nerve cells after hypoxia is the marker for converting extracellular hypoxia information to intracellular changes at hypoxia, and it is suspected that the increase of c-Fos protein can lead to the synthesis and excretion of related neurotrophic factor and nerve growth factor. However, it is still unclear what functional changes of nerve cells are induced by the increase of c-Fos protein at hypoxia, and whether it is good for the survival of damaged neurons. OBJECTIVE: To observe the expression of c-Fos in the cerebral neurons from embryos of rats with hypoxia in uterus, and investigate the pathway for the protective effect of Angelica sinensis injection on the cerebral neurons from rat embryos under hypoxia. DESIGN: A completely randomized controlled study. SETTING: Department of Histology and Embryology, Luzhou Medical College. MATERIALS: Twelve female Wistar rats in oestrum and 1 male adult Wistar rat with body mass of 220 to 250 g were selected. Rabbit-anti-rat neuro-specific enolase (NSE) and rabbit-anti-rat c-Fos were purchased from Wuhan Boster Biological Technology Co., Ltd.; Double-staining kit was bought from Beijing Zhongshan Golden Bridge Biotechnology Co., Ltd. Angelica sinensis injection was produced by the Department of Pharmacy, the Second Affiliated Hospital of Hubei Medical University. METHODS: The experiments were completed in the experimental animal center and the Department of Histology and Embryology of Luzhou Medical College from December 2004 to December 2005. ① Twelve adult female Wistar rats in oestrum and 1 male Wistar rat were housed in one rearing cage. The appearance of vaginal embolus at 8:00 in the next morning was recorded as 0 day of pregnancy and the rats were recorded for 15 days, and they were divided randomly into three groups, control group (n =4), hypoxia group (n =4) and Angelica group (n =4). BACKGROUND: Both c-Fos protein and nitricoxide synthase (NOS) have been used as general indexes in relative research about neurons, but it is lack of reports that c-Fos protein and NOS are applied synchronously to study the neurons of hypoxic fetal rats in uterus. OBJECTIVE: To study the effect of hypoxia in uterus on the expression of c-Fos protein and NOS in neurons of cerebral cortex from fetal rats and whether Angelica sinensis has the protective effect on these neurons in hypoxia. DESIGN: Randomized control experiment. SETTING: Department of Histology and Embryology, Luzhou Medical College. MATERIALS: Twelve adult female Wistar rats in oestrum and 1 male Wistar rat with bodymass from 220 to 250 g were chosen. Parenteral solution of Angelica sinensis mainly contained angelica sinensis, 10 mL/ampoule, was provided by Department of Agent of the Second Hospital Affiliated to Hubei Medical University (batch number: 01062310). METHODS: This experiment was completed in the Department of Histology and Embryology of Luzhou Medical College from September 2003 to June 2004. ① Twelve adult female Wistar rats in oestrum and 1 male Wistar rat were housed in one rearing cage. Vaginal embolus was performed on conceive female rat at 8:00 am next day. On the 15th conceiving day, all conceiving rats were divided randomly into three groups: control group, hypoxia group and Angelica group with 4 in each group. Rats in hypoxia group and Angelica group were modeled with hypotonic hypoxia in uterus. Angelica group: Rats were injected with 8 mL/kg Angelica sinensis injection through caudal veins before hypoxia. Hypoxia group: Rats were injected with the same volume of saline. Control group: Rats were not modeled and fed with normal way. ② Twenty embryos of rats were chosen randomly from each group and then routinely embedded in paraffin. 为进行耦合空间声场特性及民族音乐厅堂音质的基础声学研究,与日本神户大学安藤四一声学研究室合作,对广州南方剧院进行了双耳脉冲响应及其它主要声学参数的测试,并根据所测得的双耳脉冲响应计算得出一系列重要的厅堂客观声学指标,为进一步进行主观评价,探讨主客观指标之间相互关系以及实现可听化奠定基础。

Synaptic and blood-brain barrier structural changes in a rat epilepsy model induced by coriaria lacton Replication experiment with animals被引量:2

BACKGROUND: Structural and functional synaptic changes, as well as blood-brain barrier (BBB) changes, affect the micro-environment of nervous tissue and excitation, both of which play an important role in epilepsy. OBJECTIVE: To observe synaptic and BBB ultrastructural changes in the motor cortex of a rat epilepsy model induced by coriaria lacton, and to investigate the synaptic and BBB effects on the mechanism of epilepsy. DESIGN: A randomized controlled animal experiment. SETTING: Department of Histology and Embryology, Luzhou Medical College; and Electron Microscopy Laboratory, Luzhou Medical College. MATERIALS: Twenty healthy male Sprague Dawley rats, aged 8 weeks, were chosen for this study. The rats weighed (280 ± 50) g and were supplied by the Experimental Animal Center of Luzhou Medical College. Experimentation was performed in accordance with the ethical guidelines for the use and care of animals. The animals were randomly divided into a control group and an epilepsy group, with 10 rats in each group. METHODS: This study was performed at the Department of Histology and Embryology, and Electron Microscopy Laboratory, Luzhou Medical College between February and December 2006. According to the protocol, the epilepsy group was injected with 10 μL/100 g coriaria lacton into the lateral ventricles to establish an epileptic model. The control group rats were not administered anything. Eight days after the model was established, all rats were anesthetized with ether. The motor cortex was removed and sectioned into ultrathin sections. Synaptic and BBB ultrastructural changes were observed by electron microscopy. MAIN OUTCOME MEASURES: ①Structural changes of three different parts of the synapses, synaptic cleft width, postsynaptic density thickness, proportion of perforation synapses, curvature of synaptic interface, and length of active zones. ②Capillary and BBB changes (endothelium, basement membrane, pericyte, and the astrocyte endfeet). A Two-Lane Cellular Automata Model with Influence of Next-Nearest Neighbor Vehicle被引量:7

<正> In this paper,we propose a new two-lane cellular automata model in which the influence of the next-nearestneighbor vehicle is considered.The attributes of the traffic system composed of fast-lane and slow-lane are investigated bythe new traffic model.The simulation results show that the proposed two-lane traffic model can reproduce some trafficphenomena observed in real traffic,and that maximum flux and critical density are close to the field measurements.Moreover,the initial density distribution of the fast-lane and slow-lane has much influence on the traffic flow states.With the ratio between the densities of slow lane and fast lane increasing the lane changing frequency increases,butmaximum flux decreases.Finally,the influence of the sensitivity coefficients is discussed.

华支睾吸虫mGST新基因的克隆、表达与纯化(英文)

目的扩增华支睾吸虫一个微粒体谷胱甘肽转移酶( mGST)新基因,明确是否存在内含子,并进行原核表达与纯化。方法用PCR方法分别从华支睾吸虫成虫cDNA(质粒)文库和基因组DNA中扩增mGST基因并测序,将编码基因定向克隆到原核表达载体pET-30a( +) ,在大肠埃希菌BL21/DE3中表达,按照Ni-NTA agarose说明书纯化重组蛋白,用SDS-PAGE和TLC分析。结果mGST基因全长486 bp ,没有内含子,构建的原核表达质粒pET-30a( +)-mGST在大肠埃希菌中得到了有效表达,融合蛋白的分子质量单位约为24 ku。重组蛋白达细菌总蛋白质的11 %,纯化后的重组蛋白纯度为83 %。结论mGST基因没有内含子,在大肠埃希菌中能有效表达,得到了纯化的重组蛋白,为进一步研究其功能奠定了基础。

Experimental research on mercury emission from one-dimensional combustion test facility

The research of mercury release from coal combustion and mercury speciation in flue gas was conducted in a one-dimensional combustion test facility. The experimental results indicated that combustion temperature was the primary factor in affecting mercury vaporization and release. Experimental measurements showed high mercury levels in the particulate phase. Hg(S) is enriched in fly ash and dispersed in bottom ash. Hg(B) content decreases and the Hg^2+(g) content increases with higher furnace temperature. At 1 100℃, the levels of Hg2+(g) are 17%-48% for limited chemical kinetics .The mercury equilibrium in the flue-gas is frozen below some temperature.
